Ascertaining of Ug99-race specific genes in wheat genotypes assigned to stem rust resistance based on phenotypic and genotypic reaction CIGR Journal
Draz, Ibrahim Sobhy.
Five wheat genotypes including two commercial cultivars (Mist1 and Misr2) and three Ug99-race specific genes (Sr24, Sr25, and Sr31) were screened for stem rust resistance based on phenotypic and genotypic reaction. Based on phenotypic reaction, Sr24 (LcSr24Ag) and Sr31 (Seri-82) exhibited seeding and adult plant resistance while the wheat cultivars, Misr1 and Misr2 were susceptible. Sr25 Agatha (CI 14048)/9*LMPG-6 DK16 exhibited seedling resistance while showed susceptibility at adult plant stage. Based on genotypic reaction, DNA markers showed that gene Sr25 was only found to be present in Misr1 and Misr2 while Sr24 and Sr31 were absent in both cultivars. Although, virulence to Sr25 was detected. However, avirulence to Sr24 remains this gene as a suitable...

Assessment of EST-SSR markers for genetic analisys on coffee Bragantia
Missio,Robson Fernando; Caixeta,Eveline Teixeira; Zambolim,Eunize Maciel; Pena,Guilherme Ferreira; Ribeiro,Ana Paula; Zambolim,Laércio; Pereira,Antônio Alves; Sakiyama,Ney Sussumu.
EST-SSR markers were used to investigate the genetic diversity among and within coffee populations, to explore the possibility of their use for fingerprinting of cultivars and to assist breeding programs. Seventeen markers, developed from ESTs (Expressed Sequence Tags) from the Brazilian Coffee Genome Project, were used. All markers showed polymorphism among the genotypes assessed. The average number of allele per primer was 5.1. The highest polymorphisms were found within C. canephora (88.2%) and rust-resistant varieties (35.3%). About 29.4% of the markers differentiated C. arabica from Híbrido de Timor; it was also possible to identify those closest and farthest from C. arabica . The analysis of population-grouped genotypes revealed a 64.0% genetic...

Genetic diversity and nonparametric statistics to identify possible ISSR marker association with fiber quality of pineapple Anais da ABC (AABC)
SILVA,JULIANNA M.; LIMA,PAULO R.L.; SOUZA,FERNANDA V.D.; LEDO,CARLOS A.S.; SOUZA,EVERTON H.; PESTANA,KATIA N.; FERREIRA,CLÁUDIA F..
Abstract: Due to the increasing search for renewable resources, plant fibers have become an alternative when creating new products. Studies demonstrate the potential use of pineapple fibers in composites. The objective of this work was to evaluate the genetic diversity and verify any association between ISSR (Inter Simple Sequence Repeats) bands and quality of pineapple fibers for use in cements in the civil construction. The study analyzed the genetic variability of 11 pineapple genotypes, as well as the possible association of 131 bands from 16 ISSR markers with fiber quality characteristics. Eleven bands were selected based on their high correlations (0.64578* to 0.72457*) with three fiber quality variables. Of these, two bands were purified, sequenced,...

Genetic relationship in Coffea species and parentage determination of interspecific hybrids using ISSR (Inter- Simple Sequence Repeat) markers Genet. Mol. Biol.
Ruas,Paulo M.; Ruas,Claudete F.; Rampim,Leandro; Carvalho,Valdemar P.; Ruas,Eduardo A.; Sera,Tumoru.
Inter-simple sequence repeat (ISSR) markers were used to evaluate genetic divergence among eight Coffea species and to identify the parentage of six interspecific hybrids. A total of 14 primers which contained different simple sequence repeats (SSR) were used as single primers or combined in pairs and tested for PCR amplifications. Two hundred and thirty highly reproducible fragments were amplified, which were then used to estimate the genetic similarity and to cluster the Coffea species and hybrids. High levels of interspecific genetic variation were revealed. The dinucleotide motif (GA)9T combined with other di- tri- and tetra-nucleotides produced a greater number of DNA fragments, mostly polymorphics, suggesting a high frequency of the poly GA...

Genetic variability of pre and post-fragmentation cohorts of Aspidosperma polyneuron Muell. Arg. (Apocynaceae) BABT
Torezan,José Marcelo Domingues; Souza,Rogério Fernandes de; Ruas,Paulo Maurício; Ruas,Claudete de Fátima; Camargo,Elvis Hassmann; Vanzela,André Luís Laforga.
RAPD was used to access the genetic variability in Aspisdosperma polyneuron, a long-lived, late-reproducing tropical tree, and highly important for the Atlantic Forest. RAPD profiles from adults (pre-fragmentation, >300 years old) and seedlings (post-fragmentation, <<50 years old) were analyzed. Results showed a decrease of genetic polymorphism of post-fragmentation cohorts in small fragments and higher genetic diversity within population. The genetic diversity distribution suggested the establishment of fragments as protected reserves, and the transference of seedlings among fragments for conservation of A. polyneuron.

Background: Memecylon species are commonly used in Indian ethnomedical practices. The accurate identification is vital to enhance the drug's efficacy and biosafety. In the present study, PCR based techniques like RAPD, ISSR and DNA barcoding regions, such as 5s, psbA-trnH, rpoC1, ndh and atpF-atpH, were used to authenticate and analyze the diversity of five Memecylon species collected from Western Ghats of India. Results: Phylogenetic analysis clearly distinguished Memecylon malabaricum from Memecylon wightii and Memecylon umbellatum from Memecylon edule and clades formed are in accordance with morphological keys. In the RAPD and ISSR analyses, 27 accessions representing five Memecylon species were distinctly separated into three different clades. M....

Identification of SSR and RAPD markers linked to a resistance allele for angular leaf spot in the common bean (Phaseolus vulgaris) line ESAL 550 Genet. Mol. Biol.
Silva,Gilvan Ferreira da; Santos,João Bosco dos; Ramalho,Magno Antonio Patto.
The objective of this study was to identify RAPD and SSR markers associated with a resistant allele for angular leaf spot (Phaeoisariopsis griseola) from the line 'ESAL 550', derived from the Andean 'Jalo EEP 558' cultivar, to assist selection of resistant genotypes. The resistant line 'ESAL 550' and the susceptible cultivar 'Carioca MG' were crossed to generate F1 and F2 populations. One hundred and twenty F2:3 families were evaluated. The DNA of the 12 most resistant families was bulked and the same was done with the DNA of the 10 most susceptible, generating two contrasting bulks. One RAPD and one SSR marker was found to be linked in coupling phase to the resistant allele. The SSR marker was amplified by the primer PV-atct001(282C), and its distance...

Introgression of LTP2 gene through marker assisted backcross in barley (Hordeum vulgare L.) Electron. J. Biotechnol.
Mikolajczak,Krzysztof; Ogrodowicz,Piotr; Surma,Maria; Adamski,Tadeusz; Kuczyñska,Anetta.
Background: Marker-assisted introgression currently represents the most widely spread application of DNA markers as an aid to selection in plant breeding. New barley germplasm should be supplemented by genes that facilitate growth and development under stressful conditions. The homology search against known genes is a fundamental approach to identify genes among the generated sequences. This procedure can be utilized for SNP search in genes of predicted function of interest and associated gene ontology (GO). Results: Backcross breeding enhanced by marker selection may become a powerful method to transfer one or a few genes controlling a specific trait. In the study, the integrated approach of combining phenotypic selection with marker assisted backcross...
